I am taught "preparation of permanent slides" by one of my teachers in the course of Biological Techniques. He gave me the following steps to prepare a permanent slide:
Totally, there are seven steps in that, however the confusion lies in the fact that the step involved in dehydration is used twice. I didn’t understand that if the specimens are dehydrated once, then why is there need of dehydrating them again? Isn’t it that after the dehydrating step, the specimen is rehydrated in the staining process, then there will again be the need of dehydration process?
